Extracellular

Glycoprotein

from Virulent and Avirulent

Cryptococcus Species

Two virulent strains of Cryptococcus neoformans and two nonvirulent forms

(C. albidus and C. laurentii) weregrown in liquid cultureto produce maximal

capsule formation. A glycoproteinwasisolated from the culture medium andwas

homogeneous as determined by cellulose acetate electrophoresis and

anion-ex-changechromatography. The amino acid, neutralsugar,aminosugar,uronicacid,

and O-acetyl compositions and the infrared spectra of the glycoprotein were

determined. Theproduct of the C. neoformans strains containedmore mannose

and uronic acid than did that from the nonpathogenic strains. O-acetyl groups

wereabsent from glycoprotein of thetwononpathogens.

The main capsular polysaccharide produced

by speciesofCryptococcusisreportedtoconsist

of a mannosebackbone withside chains of xylose

andglucuronicacid (5,6,33)invarying

propor-tions.Infraredspectroscopy has shown that

dif-ferences in serological types dependin part on

thedegree ofacetylationofpolysaccharide side

chains (29). Soluble polysaccharides from

viru-lent strains are largerand contain more uronic

acid residuesthan weakly virulent strains (24).

Theproteincontentof extracellular materialhas

not been investigated, although yeasts are

known toproduce externalenzymes (27).

Quan-titiesof individual extracellularpolysaccharides

areoftenlimited,and in such casesappropriate

microanalytical proceduresareneeded(9).

Growth conditions affectthe productionand

composition of cryptococcal extracellular and

capsular material (8, 14, 15, 30,42), and media

have been developed which enhance capsule

production(30).

We report hereresultsofexperimentsin which

virulentandnonvirulentspeciesofCryptococcus

were grown in culture with maximal capsule

production. An extracellular glycoprotein was

isolated and characterized by microanalytical

procedures. The glycoprotein from C.

neofor-manscontainedO-acetylgroups and more

man-noseand uronicacidthan didthemoleculefrom

the nonpathogenic species, which did not

con-tainO-acetylgroups.

MATERIALS AND METHODS

Yeast isolates and culture conditions. C.

neo-formans strains 365-11 and 365-16 and C. laurentii

371-1 were obtained fromL. Kapica, Department of Microbiology, McGill University, Montreal, Quebec, Canada. C. albidusH1354, isolated from human cer-ebrospinal fluid, was obtained from H. G. Weiser, Aarau, Switzerland. H. J. Phaff (personal communi-cation) confirmed identificationsby expanded assimi-lation tests. The C. albidus strain was identified as a typical representativeof C. albidus var.diffluens.

Allculturesweremaintainedat4°ConSabouraud dextrose (4%) agar slopes and were subcultured monthly. Inoculum cultures of each strain were pre-pared in50ml of Littman'scapsule medium (LCM) (30)containing0.5gof dextrose whichwasadded after autoclaving. Thiamine (10 ug/ml) wasadded before autoclaving. The cultureswereshakenat 100rpmin anR77Metabolyte shakingwaterbath (New Bruns-wick Scientific Co., New Brunswick, N.J.) at 25°C until each strain reachedexponential phase.Samples (5ml) of exponential-phase culturesweretransferred to 350ml of thesamemedium in 1-literErlenmeyer flasks andwereshakenat90 rpm at25°C. The cultures were grown until they reached stationary phase, as judged by absorbance measurements and pH (ap-proaching3.0).ThepH of cultures containing patho-genic strains wasadjusted to 6.0with0.1 N NaOH, and the cells werekilled by heating the flasks in a waterbathat55°C for90min.Allcellswereharvested by centrifugationat16,000xgandwashed twice with water todislodgeasmuch adhering capsularmaterial aspossible. Washingswerecombined with theculture fluidandpassed through amembrane filter(0.45-jum poresize;MilliporeCorp., Bedford, Mass.)to remove whole cellsandcellfragments.

Virulence testing with mice. Virulence was testedbyintracerebral inoculation of12Swiss white mice per strain with a 10'; cellsuspension insterile saline(0.02to0.04ml).Deathsbeganto occurafter4 weeks. Mortalitywasexpressed astimetaken to kill 50% of the mice.Autopsieswereperformed,agarslants were inoculated with brain tissue, and smears were examined withfungicidalIndia inkby phase-contrast microscopy.

Preparationof extracellular material. The

filtered medium was by the modified procedure of Farhi et al. (14). The culture fluids were concentrated to 1/10volume by rotary evaporation at 50°C. Two volumes of absolute ethanol were added to the con-centrate,and thesolution waskept for 48 h at 4°C. Theprecipitate was collected by centrifugation at 300 xg. Onevolume of absolute ethanol was added to the supernatant, and the solution was kept for 48 h at4°C. Theprecipitate was collectedby centrifugation at 300 x g. The two precipitates were combined, washed twice with absolute ethanol, centrifuged at 300x g, and suspended indeionized water. The suspension was dialyzed at 4°C against three changes of deionized water in a 20-liter Multiple Dialyzer (Oxford Labora-tories, FosterCity, Calif.). The nondiffusible material was cleared bycentrifugation at 27,000xg for 60min at2°C, dispensed into preweighedplastic beakers, and lyophilized. The beakers were reweighed, and the white fluffymaterialwasstored inscrew-capped vials at 4°C. At least two preparations were made from eachstraingrown underidentical conditions. All ana-lytical procedureswereperformed on alcohol-precipi-tated material.

Electrophoreticprocedures. Polyacrvlamide disc electrophoresis wasperformed according to the mod-ified method ofOrnstein and Davis (37;D.J.Fox, D. A. Thurman, and D. Boulter, Biochem. J. 87:29P, 1963). Thestacking gel was omitted from the columns. Samples (0.05 to 0.2 ml containing 15 to 20 mg/ml) were run at 3 mA per tube until thebromophenol blue marker reached the bottom of the gel column. Protein was detected bystaining with 1%; amido Schwarz in 7(4 (wt/vol) aceticacid for 1 h. Destaining was done for 1 to 3 daysin7% acetic acid. Carbohydrate was detected byamodifiedperiodic acid-Schiff stain (38). Celluloseacetateelectrophoresiswasperformed in aGelmanelectrophoresis unit (Gelman Sciences, Inc., AnnArbor,Mich.). ExtracelUular material (2 mg) was dissolved in 500

of0.05 M tris(hydroxymethyl)-aminomethane barbital-sodium barbital buffer, pH 8.8 (Gelman High Resolution Barbital Buffer). Samples (5

wereappliedtothecellulose acetate strips (2.5 by15cm) andrun at 3mAperstrip for40min. The

strips were cutin halflongitudinally. One half was

stained with Alcian blueto detectglycoprotein (47); the other was stained for carboxyl groups with 1% Alcian blue in 2.5%(wt/vol)acetic acid anddestained

intwochangesof0.1Mcitratebuffer,pH 3.0-absolute

ethanol (1:1, vol/vol) and then cleared with two changes of absolute methanol (1 min each) and one rinseof 10%(vol/vol)acetic acid inabsolutemethanol (1min).Separate stripswerestainedfor protein with 0.25% Coomassie brilliant blue in 7%(wt/vol) acetic acidfor 15min (11). These strips were destained in fourchanges of 5% acetic acidandclearedasdescribed above. Thestripsweredriedat60°C for15min on a

grease-freeglass plate. Heparin (for Alcian blue)and

Hyland controlserum (for Coomassie brilliant blue) wereusedasreference markers.

Gel chromatography. A 15-mgsample was sus-pended in 10 ml of 0.02 M pyridine hydrochloride buffer, pH 5.5,and then degassed.The solution was dialyzed at 4°C against three changes of the same buffer and waschromatographedon acolumn (37by

2.5cm)ofdiethylaminoethyl (DEAE)-Bio-GelA(100

to 200mesh; Bio-Rad Laboratories, Richmond,Calif.) which wasequilibrated with the buffer. The column was eluted with 400 ml of the buffer followed by a convex gradient from 0 to3.0 M NaCl. (The mixing chamber contained700 ml ofbuffer, and thereservoir contained 700 ml of 3.0 M NaCl.) Fractions (10 ml) were collected, and portions (2 ml) were assaved for carbohydrate (10) and for protein by ultraviolet light absorbanceat 280nm.Conductivity was measured by using a conductivity bridge model RC216B2 (Indus-trial Instruments Inc., Cedar Grove, N.J.) and was proportionaltomolarity of NaCl.

Analytical procedures. Samples from stock

so-lution (2 mg of driedmaterial per ml of water) were used in analytical procedures. Analyses for nitrogen wereperformed by OrganicMicroanalyses Ltd., Mon-treal. Phosphorus wasdetermined spectrophotomet-rically (3) with KH2PO, as the standard. The ash content wasdeterminedgravimetrically after heating toconstantweightat800°C.

Amino acid composition wasdetermined after hy-drolysis invacuo of 4- to5-mgsamples with0.5 ml of 6NHCI containing oxalic acid(1mg/ml) (20) at110°C for24and48h. Thehydrolysatesweredried invacuo anddissolvedin 1.0ml of0.02N sodium citratebuffer, pH2.2.The amino acidswere analyzedwitha Beck-man amino acidanalyzer model 120C (Beckman In-struments, Inc., PaloAlto, Calif.). Basic amino acids wereseparatedon acolumn(16.5by0.9cm)(9). After the application of the sample, thespace above the resinwasfilledwithpH3.25buffer(44),andthen the column was developedat pH 5.25. This allowed the separation oflysinefrom anunknownsubstance and also improved the resolution of

tyrosine/phenylala-nineandglucosamine/galactosamine.

Amino sugars were analyzed with the amino acid analyzer afterhydrolysisin vacuowith2 NHCI(36) containing oxalic acid(1mg/ml)at105°Cfor8, 16, 24, 72,and96 h. Thehydrolysatewasanalyzed bothby themodified short-columnprocedureandby using the long-columnof the amino acidanalyzer.

Neutral sugars were determined, after hydrolysis, by gas-liquid chromatography of their trimethylsilyl derivatives. Theprocedurewasbasedon the method ofLehnhardt and Winzler (28).Samples (1 mg) were hydrolyzed withion-exchange resin in 0.02 NHCIat 100°C for 24, 48, and 72 h (35). The samples were recovered fromtheresin,driedbyrotaryevaporation, treated with0.5mlofdimethylformamidecontaining

0.1 M2-hydroxypyridine, andallowedto mutarotate

overnightat40°C (40).Thesugarswere silylatedby shakingfor 30min with0.5mlofhexamethyldisilazane and 0.25 ml oftrimethylchlorosilane. Samples (2,l) wereinjectedinto aHewlett-Packard 7610A

dual-col-umn gas chromatograph (Hewlett-Packard Co.,

Mountain View, Calif.) fitted with flame ionization detectors anddirect-on-column injection. Flow rates were:He,60ml/min; H2,35ml/min;andair,500ml/ min.Dual copper columns(2.4mby6mm)contained 10%(wt/wt)SE-52onDiaport S(80to100mesh),and the chromatograph was operated isothermally at 190°Cwith a flash heatertemperatureof260°Canda detectortemperatureof270°C.Molarresponsefactors foreachsugarrelative tomannitol (12) and the per-centagecompositionofeach of thesugarsat

tationequilibriumweredetermined. Uronic acidswere estimated colorimetrically with metaphenylphenol, usingglucuronolactone asthe standard (7).

The method of O-acetyl determination involved transesterification with sodium methoxide (0.1 M in absolutemethanol)at0°C for30min(48). The result-ingmethyl acetate wasestimatedby gas chromatog-raphy (P. E.Reid, personalcommunication)ina Hew-lett-Packard F&M402gaschromatograph fittedwith flame ionization detectors.

Infrared spectra were obtained by using approxi-mately1.5mg ofdry material whichwasgroundwith 200 mgof dryKBr, compressedat 18tons(19.8 metric tons),and scanned inaUnicamSP 200G spectropho-tometer(Pye UnicamLtd.,Cambridge,England).

RESULTS

Growth patterns. All strains grew well at

25°C, and exponential phase was established

after 36 h. The pH optima for cell growth and subsequent capsule production for all five strains

were between pH 6.5 and 7.0 at inoculation,

although capsular material and visible capsules

did not appear in the medium or on the cells

until thepHdroppedtobelow 5.1. Thecapsule

thicknessranged from 1.5 to 4.0tIm.

Virulencetests.Both C. albidusand C.

lau-rentii failed to kill any ofthe mice. C.

neofor-mans 365-11killed50%ofthe miceafter5days,

and C.neoformans365-16caused50%mortality

after 18days.

Electrophoresis of extracellular

mate-rial.Polyacrylamideelectrophoresis patternsof

theextracellularmaterialsrevealedcongruence

of periodic acid-Schiff and amido Schwarz

bands,indicatingthatthere wasonly one major

glycoprotein. Cellulose acetate electrophoresis

revealed asingle periodic acid-Schiff and Alcian

bluepositiveband in eachpreparation. FromC.

albidus this band hadamobility of0.28relative

to heparin, fromC. Iaurentiithe value was0.48,

andfrom C.neoformansthe valuewasbetween

0.31 and 0.34. The viscosity of the material in

solution imposed limits uponloading for

electro-phoresis.

Gelchromatography. The fractionation of

extracellular material on a DEAE-Bio-Gel A

column produced asingle, sharply defined band

which elutedatthe start of the NaCl gradient.

ThepatternforC. neoformans365-11 isshown

in Fig. 1. Protein and carbohydrate moieties

werecoincident within this band. This

observa-tion, together with the electrophoretic results,

suggests that a small protein fraction may be

bound to a large acidic carbohydrate moiety.

There did not appear to be contamination by

othermolecularspecies.

N, P, and ashanalyses. Thetotal N inany

preparation was between 0.1 and 0.2% by weight.

The recovery of N calculated from amino acid

analyseswas between 40and 100% of the total

N analysis. The total P was between 0.1 and

0.2%

except

forthe

C.

neoformans strains, where

the P content was 0.02 to 0.4%. The ash contents

varied substantially among preparations but

usuallywerebetween 0.2 and 0.1%.

Amino acid and amino sugar

composi-tion. The commonest aminoacidswere

aspar-tate/asparagine (Asx), glutamate/glutamine

(Glx),serine, and threonine. C.

neoformans

365-16 had large amounts of proline and differed

substantially in the ratios of glycine/alanine,

isoleucine/leucine, and

tyrosine/phenylalanine

from the other strains and species. C.

pathogenic species although it had a large

amountof Asx. The extracellularmaterial

con-tained lesseramountsoftotal basic amino acids

than did cell walls (0.1 and 0.2%, respectively)

and also more serine and threonine (0.7 and

0.1%, respectively). Analysis of all the strains

showedashoulder emerging justahead of lysine,

and C. albidus gave a clearly resolved peak

before lysine. Spectrophotometric tests for

hy-droxyprolinewere allnegative. Glucosaminewas

detected inhydrolysatesofallpreparations

(Ta-ble 1), butnogalactosaminewasdetectedinthe

strains examined.

Neutral sugars. Table 2 shows results of

neutral sugar analyses. The same four sugars

were detected in all strains, although the

pro-portions varied substantially. Mannosewasthe

major constituentinall samples. Total

recover-ies were approximately 30% higher after resin

hydrolysis than after hydrolysis using 2 N

tri-fluoroaceticacid(2, 9).

Uronic acids and O-acetylgroups. Table

3 presents results of uronic acid and O-acetyl

groupanalyses. The C. neoformans strains

con-tainedmuchmoreuronicacid thandid the

non-pathogenic species. O-acetyl groups were

de-tected only in C. neoformans. Strain 365-11, whichhada.higher50%lethal doseinourtests,

(1 mg/ml) for 24, 48, and 72 h. For amino sugars,

duplicate samples (4 to 5 mg/ml) were hydrolyzed

with2N HClcontaining oxalic acid (1mg/ml) for 8, 16,24, 72, and 96 h.

hadsubstantiallymoreO-acetylationthan strain

365-16. These results indicate that 35% of the

total polysaccharide was O-acetylated in the

extracellular material from C. neoformans

365-11. The value for 365-16 was 7%. We did not

determine where the O-acetyl groups were

sub-stitutedorin which sugars they occurred.

Complete analysis of the extracellular

material. Table3summarizestotalrecovery of

the extracellular material components for the

four Cryptococcus strains. Absolute recoveries

rangedfrom99.2 to131.7% byweight.We cannot

account for this major discrepancyin absolute

recovery.Lipid determinationsare notincluded

because gravimetric methods werefoundto be

unreliableonthesmallamountsavailable.

How-ever, thevalues obtained were never less than

3%by weight.

Infrared

spectroscopy. Material from the

four strains showed identical spectral patterns

forwavenumbers between2,000 and 4,000cm-'

(Fig. 2). Authentic mannan showed the same

absorption maxima at2,900 and 3,250 to 3,550

cm-'

(19). Patterns were also similar between

650 and 1,800

cm-'

(Fig. 3), but there were

differences in intensity at 1,730, 1,650, 1,550,

1,375, 1,250, and 800 cm-'. Material from C.

laurentii andastandardfor glucosamine

hydro-chloridelacked the intense peak at 1,730

cm-'

caused by carbonyl stretching vibrations. This

peakwaspresent in the C.neoformans strains.

C.laurentii and C. albidus showed vibrationsat

1,550 and 1,620 to 1,635

cm-'.

This correlated

with largeramounts ofaminoacids and

gluco-saminerecovered from thesestrains (Table 3).

Thesignalis attributed toC,0,and NH groups

(4).The increasedintensityof vibrationsat 1,250

cm-' and 1,730

cm-'

(Fig. 3) correlated with

increased acetylation and uronic acid content

(Table3). Asimilar slightincrease inmethylH

at 1,375

cm-'

and in carboxylateanion at 1,425

cm' also confirmedtheincreaseinO-acetyland

carboxylic acid present. The more virulent C.

neoformans 365-11 showed the more intense

bandsatallofthesewavenumbers.

DISCUSSION

Standard culture conditionswereadopted for

maximalcapsuleproductioninLCM.Theywere

nottheconditionsassumedtooccurinvivo,but

were selectedsothatcomparative experiments

and analytical procedures could be performed

onmaterialsproducedunderidentical, and thus

comparable,

culture conditions. A temperature

of25°Cwaschosen becausetheC. albidusstrain

didnotgrow at

370C.

sizewas obtained on solid LCMatpH 7.5and

withthe fact that C.

neofornans

must be able

to grow atpH7.34(spinal fluid)and 7.39(blood

plasma) invivo. We confirmed other reports(1,

15) thatlarge amountsofheteropolysaccharide

wereproduced by cells inoculated in LCMat an

initialpHof 7.0.Recoveries of extracellular

ma-terialwerepoor whencells were harvested

be-forethey reached stationary phase. Wedid not

determine thecauseof thepH droptoward pH

3.0inthisphase. After5daysofincubation, the

C.

neoforrnans

culturesweremuchmore

muci-laginous and

slimy

than those of the

nonpatho-gens, and the ethanol

precipitates

from culture

fluidwere morefibrous. The increased

viscosity

seemed to be accompanied by increases in

0-acetyl

and uronic acidcontents

(Table

3).

Columnchromatography (Fig. 1) and

electro-phoretic analysis

followed

by various tests for

carbohydrate, protein, and uronicacid indicated

that the

extracellular

material behaved as a

single molecular species. Ourtestsfor

homoge-neitydid notrevealthepresence of any

contam-inating materials.

Theproportionsofmannose,xylose (Table2),

and uronic acid (Table 3) in our preparations

weresimilar to those in other reports (6, 14, 18,

43) but different from results obtained for C.

neoformans

serotype B (5),whichwas

recently

reclassified as C. bacillisporus (26). The

pres-enceofgalactose (whichwe found in all of our

ers (13, 39) found two major capsular

polysac-charides, only one ofwhichcontainedgalactose.

Golubevetal. (16) reported thatcapsular

poly-saccharides contained galactose, whereas those

from the extracellular material did not. Still

others (14) foundnodifference in thegalactose

contentof adhered and solublepolysaccharides.

Our preparationswerederivedfromsoluble and

easily released capsular materials. Our detection

ofglucoseisthe firstfor thissugar as a

compo-nent of cryptococcal extracellular

polysaccha-rides.

The major differences between the strains

were in uronic acid content, first reported by

Kozel and Cazin (24), and in the degree of

0-acetyl substitution. These differences were

clearlyshown inthe infraredspectra (Fig.3) in

spite of the difficulties in interpretation which

often arise because ofthe additive effects and

influences of the absorptions of the different

chemicalgroups present. The

peak

at1,250cm-'

is undoubtedly duetoC-O stretching vibrations,

but the line betweenthe esterfunctionat 1,725

to 1,750cm-' andthecarboxylat 1,650 to 1,725

cm ' is hard to distinguish, especially if both

functions areknown to be present. It has been

found(17, 46) thattheesterfunction, presumed

tobe O-acetyl, is hydrolytically cleaved under

mild alkaline conditions withresulting lossesof

absorption at 1,725 and 1,250 cm-'. The

non-pathogenic species did not contain O-acetyl

functions, but did have uronic acid. It is thus

possible that the vibrations at 1,730cm-' were

duetobothacetyl andcarboxyl functions.This

is consistent with the increase in peak size at

thiswavenumberto amaximumfor C.

neofor-mans365-11.If theeffect isadditive, it explains

why the differenceinpeak sizebetweenthe two

strainsofC. neoformanswas not asmarked as

would have been expected from differences in

their

0-acetyl

contentalone. None ofthe strains

showed a carboxyl salt peak at 1,600cm-'.

0-acetyl

groups have been

reported

from C.

lau-rentiigrown on low-Pmedium(42,44) andfrom

C.bacillisporus serotype B (5).

Thehigher glucosaminecontentof C. albidus

and theC.laurentiistrain (Table1) is apparent

from the spectral enhancement at 1,550 cm-'

(Fig. 3). Theslightshoulder at 1,730

cm-'

onthe

C. albidus spectrum may have been dueto

N-acetylation. Glucosamine has not previously

been detected incryptococcalextracellular

poly-saccharides or capsules, although otheryeasts

and both yeast mycelial

phases

of

Sporothrix

schenckii produced extracellular

polysaccha-rides which contained glucosamine and

N-ace-tylglucosamine(21, 45). All of the strains showed

peaks at 800and 900cm-' representative of

a-linked glycan. The vibrations, aswell as

vibra-INFECT. IMMUN. tions at 980 cm-', were present in the yeast

mannanbut notin the yeastglucan (4).

This study is the first report of amino acid

analyses from cryptococcal extracellular poly-saccharide. From 20 to 100 tg of amino acids was recovered per mg of extracellular material

for all thestrains, and all contained at least 0.1%

(by weight) nitrogen (Table 2). Most authors

have regarded the N component as a

contami-nant. They deproteinized by extraction with

chloroform-butanol (25) during purification (14,

24, 34). This procedure left about 1.7% (by

weight) protein in their preparations, and there

may have been more in the original extracellular

materialbecausethey used 3% chloroform or 1%

phenol, the latter of which reduces amino acid

recoveries (A. Ross, Ph.D. thesis, University of

BritishColumbia, Vancouver, 1975) to kill the

cells before polysaccharide extraction.

Threo-nine, serine, and Glx were the predominant amino acids in C. albidus, whereas C.

neofor-mans 365-11 hadlargeamounts ofAsx, serine,

andGlx (Table1). InC.neoformans 365-16, Asx

and serine were the predominant amino acids

and levels of proline and phenylalanine were

higherthan in the other strain. The C.Iaurentii

hadlargeamountsof Asx and Glx. The

discrep-ancies in N recoveries suggest that there were

some losses in amino acids and amino sugars.

Thepresence oflargequantities of

polysaccha-ride causeshumin production during hydrolysis

(20), and it hasbeen shown that some losses of

amino acidsare due to adsorption to the glass

during desiccation (41).

Ouruseof virulencetestingwaslimitedto the

confirmation ofpathogenic activity in the

cul-tureswhichwereusedtoinoculatefor

glycopro-tein production. C. neoformans strains were

pathogenictotheSwisswhite mice used in our

tests, although 50%lethal dose values differed

fromthose obtainedwith other strains ofmice.

Our resultssupportpreviousreports (17, 29) that

reactivity of different serotypes of C.

neofor-mansdependsto some extent onthedegree of

acetylation. It is well established (25, 31) that

serum opsonizing agents (immunoglobulin G)

react with capsular polysaccharide to destroy

the pathogenic function (23). Kozel and Cazin

(24) found thatinjectionofcapsular

polysaccha-ride fromavirulent strain of C.neoformansdid

not inhibitphagocytosisinvitro ofa

nonencap-sulated strainorof C.laurentii.

C.albidus,the

cerebrospinal

fluid

isolate,

was

notvirulentinour tests.Itdidnotgrowat

37°C

and was not

acetylated.

It did have a

large

amount ofgalactose

(like

365-16) and

glucosa-mine (like365-11).

Table 3 shows that total recoveriesare

unre-liable measures ofsuccessin

complete

analysis.

Complete (90 to 100%) recovery of cell wall

constituents has been achieved (9). The removal ofwaterbefore analysis proved difficult in that

study. Theextracellular materials reported here

were difficult todry to constantweight before

analysis. We believe that the results reflect

fail-ure to dry (heat at 800C

overnight,

cool over

P205, and weigh 20s after removal from

desic-cator) the sample. The paradox is that different

batches of materialgavesimilar results and the

errorwas thusconstant.Clearly the matter

re-quires further study. It should be noted

that the conclusions reached from this study

remain the same if all data are calculated to

100%recovery.
