Introduction

Images acquired on the Operetta® High Content Imaging System are stored as TIF files in the Operetta Database (ODA). The ODA also contains all image-related metadata as well as image analysis sequences and results that have been generated with the Harmony® High Content Imaging and Analysis Software. There are several different ways to migrate images and the associated metadata from the Harmony software to the Volocity® 3D Image Analysis Software. Here, we provide detailed step-by-step instructions for all possible methods.

Data migration by drag and drop

The easiest way to migrate data to the Volocity software is by using the Harmony software’s drag and drop functionality. With a couple of additional steps, the drag and drop approach can also be used for data migration even if Harmony and Volocity software are not run on the same PC. In general, drag and drop allows a specific subset of wells, fields, time points and stack planes to be selected and transferred to the Volocity software.

Situation A: Harmony software and Volocity software are run on the same PC

1. Start the Harmony software and load the measurement that is to be transferred. In the Navigation Area select all wells, image fields, stack planes and time points to be transferred to the Volocity software.

2. Start the Volocity software. Drag one of the selected wells from the Navigationarea of the Harmony software and drop it into the library of the Volocity software. The Volocity software will copy all the images and image-related metadata from the ODA into the Volocity software library.

Situation B: Harmony software and Volocity software are not run on the same PC

Please note that you can only use this method if your Volocity software can establish a network connection to the PC running the ODA, which is usually the instrument PC.

1. Start the Harmony software and load the measurement that is to be transferred. In the Navigation Area select all wells, image fields, stack planes and time points to be transferred to the Volocity software.

2. Start the WordPad software, which is delivered with every Windows® installation. Drag one of the selected wells from the Navigationarea of the Harmony software and drop it into the text area of WordPad. The WordPad document will be filled with XML code. Save the document as

<FILENAME>.xml, e.g. to a USB drive.

3. On the second computer start the Volocity software and drag and drop the XML file from the Windows® Explorer® into the Volocity software library. Alternatively, you can open the XML via the

Import function from the File menu of the Volocity software. The Volocity software will copy all the images and image-related metadata from the ODA into the Volocity software library.

Data migration via export and import

Data migration via export and import will copy all images and image-related metadata from an entire plate into the library of the Volocity software. Please note that transferring the data of an entire plate to the Volocity software may be time-consuming.

1. Start the Harmony software and open the Settings window. In the Harmony Settings window open the Data Management window.

2. In the Data Management window open the Export Data dialog. There are two export methods suitable for the data migration to the Volocity software.

a. Specify Measurement Index and Referenced Images as the method for image export if your installation of the Volocity software can establish a network connection to the PC running the ODA, which is usually the instrument PC. Next, specify an export path (e.g. a USB drive) and start the export process.

b. 3. Start the Volocity software and Windows® Explorer®, and navigate to the export folder specified in step 2. There you will find a subfolder named according to the exported measurement. In the subfolder, you can find an Images folder that contains an XML file. If step 2b was selected (Measurement Index and Images) the Images folder will also contain all the exported TIF files.

4. Drag and drop the XML file from Windows® Explorer® into the library of the Volocity software.

Alternatively, you can open the XML via the Import function from the File menu of the Volocity software. The Volocity software will copy all the images and image-related metadata into its library.
