Regulation of neural enhancer activity by Ascl1 and Sox factors

Regulation of neural enhancer activity by Ascl1 and Sox factors

Mutant and wt enhancers were cotransfected in NS5 cells together with expression vectors for Ascl1 and Sox2 overexpressed either alone or simultaneously (as shown in the legend of the graphs). Luciferase activity was measured after 24 h. For each enhancer, luciferase activity was normalised to the activity of the same enhancer after overexpression of GFP as control. For each enhancer, luciferase activity of the wt element is shown on grey background in the graphs. Data are presented as the mean ± SD of triplicate assays. Panels B, D, G, I, K J represent diagram of each enhancer as in Figure 4.1 and the sequence of the motifs within each element, as explained in Figure 4.2. (A) Mutation of the E-box abolished the activation of enhancer MSB4 by exogenous Ascl1 (as shown by the black arrow in the graph). Mutation of the Sox motif did not change the activity by exogenous Sox2. Mutation of the Sox motif also abolished activation of the enhancer by exogenous Ascl1 (mutant element M2 vs wt) as shown by the red arrow in the graph. The experiment shown for enhancer MSB4 is representative of three independent biological replicates. (C) Mutagenesis of enhancer MSB18 shows that different binding motifs give different contribution to the regulation of the enhancer. Mutation of the E-box 1 abolished Ascl1-induced activation of this neuronal enhancer (mutant element M1 vs wt, as indicated by the arrow in the graph). Mutation of the Sox motif 1 did not change the activity by exogenous Sox2, which did not show activation of the enhancer. Mutation of the E-box 2, E-box 3 and Sox motif 2 (mutant elements M2, M3, and M5 vs wt element) increased strongly the activation of the enhancer by exogenous Ascl1 and also by exogenous Sox2 in mutant M5 (as indicated by the three black arrows in the graph) when Ascl1 was overexpressed alone or together with Sox2

NCLB and School Accountability the statewide test is MCAS — the Massachusetts Comprehensive Assess- ment System. Public schools in Massachusetts measure school improvement based on the school’s progress toward the goal of cutting proficiency gaps in half by 2017. Proficiency gaps are the differences among groups of students scoring at the “proficient” level on MCAS. The groups are by race/ethnicity, English language learners, students with disabilities, and low income. Stu- dents who score at the “proficient” level demonstrate a solid understanding of challenging subject matter and can solve a wide variety of problems. Based on their MCAS results, districts and schools are placed in one of five “accountability and assistance levels.” Level 1 schools are the highest performing schools. Level 3 and 4 schools are in the bottom 20% in school performance. These schools receive additional supports, which may include a longer school day and free after-school tutoring in some schools, to help them improve. Boston has no Level 5 schools.

Boston: the Red Sox, the Celtics, and Race, 1945-1969

Boston: the Red Sox, the Celtics, and Race, 1945-1969

Despite, the racism that continued to linger over the franchise, the Red Sox acted as a safe haven from the racial strife that engulfed the city during the busing crisis. Busing did not affect any of the Red Sox players in any direct way, most lived in the suburbs and were not from the city, and yet the controversy surrounded the city that they represented. The Red Sox during their runs in ’75 and ’78 featured Jim Rice, whom many consider to be the Red Sox’s first black superstar, and Luis Tiant, a black Cuban, received cheers from the Fenway faithful that were normally reserved for the team’s white stars. Ironically, oftentimes the same fans who cheered for the black players on the Red Sox were the same people who did not want African Americans going to school with their children. On this paradox Bill Reynolds writes, “As if sports and the Red Sox were somehow exempt from the hatred and tension that ran through Boston, the one island of refuge in an ever-churning sea, even if race always had been the elephant in the living room for the Red Sox, the legacy of being the last team in baseball to integrate.” 238

imposed on the corporate governance and financial reporting of public companies. For example, the passage of SOX led to the creation of the Public Company Accounting Oversight Board (PCAOB), which serves as the auditor of the auditors. This legislation requires public accounting firms that audit publicly traded companies to be registered with the PCAOB, and to subject themselves to inspections on a regular basis. Any firm auditing more than 100 publicly traded client firms must be inspected annually, while firms not meeting the 100-audits threshold are subject to inspection every three years. PCAOB, as a non-profit organization reporting to the SEC, claims to represent and protect the interests of the shareholders. By committing to inspect the auditors, the Board creates an extra tool for shareholders to reduce agency problems.

Sale struck out a season-high 13 while allowing three hits and two hits in seven innings, his third consecutive game with double-digit strikeouts and fourth in his last five. Sale struck out 12 Baltimore Orioles in his previous start last Thursday at Camden Yards, that one following a 10-strikeout effort against the Minnesota Twins on May 23 at U.S. Cellular Field. “Any time you go out there and you feel like you get in a rhythm, that’s all pitching really is is finding a groove, getting in a rhythm, just trying to — I wouldn’t say be comfortable — but find the groove and ride it as long as you can,” Sale said. “They don’t last forever.”

Audit fee discounting in the post-SOX environment

Audit fee discounting in the post-SOX environment

Despite the continued delays of SOX 404(b) compliance for non-accelerated fi lers, there was little if any anticipation of the exemption by auditors and their clients. In October 2009, the fi rst evidence of a possible permanent exemption was mentioned in the business press: on October 8, 2009, the Small Business SOX Compliance Relief Act was introduced to Congress by Representative Scott Garrett (H.R. 3775, 2009), which proposed the permanent exemption. Based on a comprehensive search, there was no mention of the possibility of a permanent exemption prior to this date[2]. This fact allows us to consider the possibility that auditors offered discounted audit pricing to their non-accelerated clients in anticipation of the signi fi cant additional fees they would earn when SOX 404(b) compliance was required. Auditors could accurately anticipate the future additional SOX 404(b) fees due to the work already being performed for their accelerated- fi ler clients. Prior research documents signi fi cant audit fee increases (i.e. from 86 to 359 per cent) for accelerated fi lers from the pre- to post-SOX 404(b) requirement periods (Raghunandan and Rama, 2006; Krishnan et al., 2008).

Firm Risk and Proxy Fights: Evidence from SOX

Firm Risk and Proxy Fights: Evidence from SOX

Second, we find that the coefficients of the High Fight Threat are significantly positive in increasing firm leverage ratio (2.17) during pre-SOX years, but other firm policies are not significantly affected by the high proxy threat before 2002. More important is the interactive term between the SOX and High Fight Threat in analyzing the disciplinary role of proxy contest during the post-SOX period. We find significant changes in at least half of our tested corporate policies. Under a high possibility of future proxy contests, firms increase their leverages, pay more dividends, and conduct more share repurchases. These policy changes reflect an improved use of the cash reserves and are largely along the direction favored by active investors, fitting well with other findings in the literature (Berger et al., 1997, La Porta et al., 2000, and Safieddine & Titman, 1999).

Simulation of SOx Concentration of Dhaka City by Air Model

Simulation of SOx Concentration of Dhaka City by Air Model

contributions to overall fine particulate emissions are- two-stroke engine three-wheelers and heavy-duty diesel vehicles. A large number of pedestrians, drivers, passengers, traffic policemen, street vendors and other groups undoubtedly suffer from significant health damage. As a result of exposure to emissions from a large variety of motorized vehicles including two-stroke auto rickshaws or "baby taxis", trucks, buses, cars and two-wheelers. They are responsible for 25% of the particulate matter and 60% of the toxic and smog-forming hydrocarbons contributed by all motor vehicles. Available air quality monitoring data suggests that the ambient concentration of some pollutants especially suspended particulate matter (SPM), and SOx in air of Dhaka. Thus the increasing contribution of atmospheric loads of SO2 and SOx to global climate change is anticipated and it is really necessary to quantify these emissions in a hurried manner. A national course- plotting committee should establish with local and expatriate Bangladeshi experts to deal with the problem of emission. Government has taken a number of initiatives including banning of polythene which had been blamed for a number of environmental hazards included water contamination, closing of drains and sewerage lines and disposal of polythene everywhere was choking the sewage, creating air pollution as leaded fuel and banning of two-stroke three-wheeler from Dhaka. Pollution control board requires performing environmental impact assessment before certifying a new industry.

Assessment of SOX implementation - from an Internal Audit perspective

Assessment of SOX implementation - from an Internal Audit perspective

Both companies utilize the Internal Audit department for their SOX implementation process. However, for the non-accelerated filer, an Internal Audit charter adoption has been delayed and SOX compliance team reports only to the CFO. Since the company is not in its compliance year yet, this is not a direct risk, however they may experience concerns with this structure from the external auditors when it comes to the reliability of the results of the internal testing. If the Internal Audit and SOX compliance is not independent from management, it may be reluctant to report any issues that concern the Senior Manager since they are reviewing the audit results and a conflict of interest arises. Anand (2006) points out that to have autonomy and opportunity to report honestly the Internal Auditors “should not be under any undue influence from executives” (p. 37). The accelerated filer on the other hand is utilizing its Internal Audit department in an effective manner and reports directly to the Audit Committee as well as to Senior Management. They utilize the Internal Auditors for the SOX compliance testing, physical inventory and financial audits. To segregate the process of SOX documentation and testing, they have created two teams that deal with SOX. This division helps ensure the objectivity of the review of the internal controls. One team suggests and implements controls and another

hypertrophic chondrocytes, exhibiting an expression profile similar to that of the type II collagen gene Col2a1 (Ng et al., 1997; Zhao et al., 1997), and it is essential throughout these developmental stages (Dy et al., 2012). Sox9 binding to the Col2a1 intronic enhancer is required for the activation of the Col2a1 gene (Bell et al., 1997; Lefebvre et al., 1997), but strong activation of the 48 bp Col2a1 minimal enhancer occurs only when Sox9 and the SoxD proteins Sox5 and Sox6 (Sox5/6) together bind to several sites in the enhancer (Lefebvre et al., 1998). The same combination of Sox9 and Sox5/6, known as the ‘Sox trio’ is involved in the activation of many extracellular matrix genes, e.g. aggrecan (Acan) and Col11a2, which are secreted by the chondrocytes (Han and Lefebvre, 2008). This ‘Sox trio’ primarily functions as combination of a Sox9 homodimer and multiple Sox5/6 dimers that bind to adjacent DNA sites (Fig. 4D). Indeed, individuals with mutations in the SOX9 dimerization domain develop campomelic dysplasia without causing sex reversal (Bernard et al., 2003; Sock et al., 2003). In line with this, single knockouts of Sox5 or Sox6 in mice lead to mild skeletal abnormalities, whereas a Sox5/6 double knockout leads to a lack of chondrogenesis despite normal Sox9 expression (Smits et al., 2001), thereby confirming the functional cooperation of SoxD and SoxE proteins during chondrogenesis. The molecular mechanism underlying the SoxD-SoxE cooperation has not been clarified.

Expression of Sox family genes in early lamprey development

A SoxD homologue was identified by bioinformatic survey of the lam- prey genomic sequences and cloned using RACE. RACE was also used to obtain full-length sequences of the Sox genes, where cDNA clones were incomplete. Total RNA was extracted from 6, 8, 10, and 14 day old embryos from Ambion:RNAquous kit. RLM-5’ RACE was conducted on the total mRNA in accordance with Invitrogen: GeneRacer Kit. Total RNA was dephosphorylated through Calf Intestinal Phosphatase (CIP) treat- ment, decapped via Tobacco Acid Pyrophosphatase (TAP), ligated with the GeneRacer RNA oligo, and finally reverse transcribed using random hexamer priming to form the cDNA template. Gene specific primers were:
