Gene transcription, regulation and the microenvironments surrounding extracellular matrix proteins play an important role in all living cells; enabling them to respond to diverse external
stimuli and to coordinate their inherent cellular programs. By engineering human transcription factor (TF) proteins tagged with a cell membrane penetration domain (CPD), such as 11 arginine (11R tag), LD Biopharma are able to deliver them directly into cells by simply introducing them into culture medium. They can also be efficiently delivered intracellularly using a cationic lipid-mediated system, such as their ProFectin reagent (Catalogue No. R-2000).
Systematically engineering human matrix proteins using refolding technology has allowed them to develop a comprehensive coating/culture system to potentially control stem cell differentiation and many cell lineages in vitroand even in vivo.

AD is a devastating neurological disease that affects more than 37 million people worldwide (1). PD is the second most common neurodegenerative disorder and affects approximately seven million people globally (2,3). The worldwide prevalence of HD is 5–10 cases per 100,000 persons (4,5). As research progresses, many similarities appear which relate these diseases to one another on a sub-cellular level. There are many parallels between different neurodegenerative disorders including atypical protein assemblies as well as induced cell death.(6,7).
Neurodegeneration can be found in many different levels of neuronal circuitry ranging from molecular to systemic. The causes and effects of neuronal degeneration are of major interest to a wide variety of neuroscientists. Paralleling this growing interest is an increasing number of methods applicable to the detection of neuronal degeneration. Biosensis offer Fluoro-Jade®C a RTD™
(Ready-to-Dilute) kit designed to stain for neuronal degeneration, with a fluorescent Nissl counter stain, regardless of specific insult or mechanism of cell death. Fluoro-Jade C exhibits great signal to background noise ratio, as well as high resolution. This translates to a stain of maximal contrast and affinity for degenerating neurons. This makes it ideal for localising not only degenerating nerve cell bodies but also distal dendrites, axons and terminals. The dye is highly resistant to fading and is compatible with virtually all histological processing and staining protocols.
The Fluoro-Jade®C 'Ready to Dilute' (RTD™)
Staining Kit provides an easy to use assortment of Fluoro-Jade C, DAPI, sodium
hydroxide and potassium permanganate in liquid form. Fluoro-Jade C labelled degenerating neurons are visualised with blue light excitation while DAPI counter stained cell nuclei are visualised with ultra-violet illumination. The Fluoro-Jade C Staining Kit can be used on all kinds of preserved tissues, including fresh-frozen, paraformaldehyde or formalin fixed, and formalin fixed, paraffin-embedded tissues.

Proteins often aggregate when exposed to elevated temperatures, long storage periods, repeated freeze/thaw cycles, to high shear forces or intense light. Oxidation by air, reactive reagents or surface exposure provide further sources of protein aggregation.
Protein aggregation can often be avoided with proper choice of pH, salt or
supplementation of stabilising additives in the buffer. This OptiSol protein
solubilisation screening kit contains a systematically varied array of buffers (from pH3 to pH10) and a series of solubility enhancers (salts, amino acids, sugars, polyols, reducing reagents) that enable the determination of conditions under which a particular protein sample is protected from aggregation. Similarly, proteins may be deaggregated by employing such formulations. A total of 90 different formulations with solubility enhancers can be tested in one single, label-free
experiment; included are 6 conditions that function as positive and negative control experiments.
Soluble proteins pass through filters, aggregated proteins don’t. This simple feature is exploited in the filtration step of the OptiSol assay. Those wells that contain soluble proteins will yield protein after the filtration process; those with protein aggregates will not contain any protein.

The system, developed by System Biosciences (SBI) uses a nuclease, CRISPR-associated (Cas9) that complexes with small RNA’s as guides (gRNAs) to cleave DNA in a sequence-specific manner upstream of the Protospacer Adaptor Motif (PAM) in any genomic location. The CRISPR-Cas9 system is comprised of separate guide RNAs known as the crRNA and tracrRNA. These two separate RNAs have been combined into a single RNA to enable site-specific mammalian genome cutting through the design of a short guide RNA. SBI has built a series of all-in-one CRISPR-Cas9 cloning vectors with different promoter options. These cloning vectors will express a human codon-optimised Cas9 nuclease as well as any gRNA
sequence of your choice linked to the tracrRNA. Simply design the gRNA for the genome target DNA sequence and clone into the Cas9 SmartNuclease™ vector for efficient genome engineering.
How to build a Cas9 SmartNuclease™ Vector
The general workflow of the cloning, validation, and transfection of the
gRNA/Cas9 expression construct into cells is simple. First, design two DNA
oligonucleotides that are sense and antisense sequences of the target DNA, which is 20 bp upstream of the Protospacer Adaptor Motif PAN (5’ – NGG – 3’) (Figure 1). Anneal the two oligonucleotides to
generate a duplex and clone the duplex directly into the provided linearised CRISPR-Cas9 vector by ligation (Figure 2). Transform into competent cells and grow
the clones using LB/Kanamycin. Confirm the correct clones by plasmid construct sequencing.

HepatoChem™ Technology
Their innovative approach is based on a diverse set of chemical conditions that mimics the main reactions occurring during drug metabolism. This panel is used to quickly find reaction conditions that
generate specific drug metabolites. This approach (basically a chemical liver), is analogous to the panels of cytochromes P450 present in the human liver, but with the advantage of the scalability of chemistry. When a reaction condition is identified, it is optimised and scaled up to produce milligram to gram quantities of metabolites.
HepatoChem™ has developed a series of biomimetic panels. These panels use several kinds of catalysts, including metalloporphyrins, which gives access to a unique set of reaction conditions. This series of easy-to-use kits is suitable for the preparation of metabolites for MS and NMR.

Antigen detection with primary antibodies of the same species as the test tissue yields high background when using an indirect detection method. This severely limits the use of mouse monoclonal antibodies on mouse tissues. Mouse Antibody On Mouse Tissue
The Klear Mouse HRP-Polymer Detection System is designed for staining mouse antibodies on mouse tissues. Mouse Antibody On Rat Tissue
Detecting mouse primary antibody on rat tissue is a very difficult task due to background staining issues. Polink-2 Plus HRP Mouse-NR (no cross reactivity to rat) DAB Detection Kit is specially designed to solve this problem. The secondary antibody is adsorbed to rat, rabbit and human serum proteins, providing excellent specificity to detect mouse primary antibody on rat tissue. Besides rat tissue Polink-2 Plus HRP Mouse-NR DAB Detection kit also can be used on human tissue and rabbit tissue. Polink-2 Plus HRP Mouse-NR DAB Detection Kit is the 3rdgeneration of polymer
detection system. It uses mouse antibody enhancer to help amplify the polymer-enzyme conjugate reaction to achieve super sensitivity and specificity in IHC staining. It produces consistent immunostaining outcomes on archival tissues and on difficult-to-work with antibodies. EdU / EU CLICK Reagents – an alternative to BrdU/BrU
Traditionally, incorporation of 5-Bromo (deoxy) uridine (5-BrdU/5-BrU) into nascent DNA or RNA strands has been employed to assess proliferation. This method is hampered by the cell morphology destructive detection via a Br(d)U specific antibody that requires harsh cell
permeabilisation conditions and nucleic acid denaturation procedures preventing subsequent analysis of other cellular targets.
This drawback can be overcome by the incorporation of ethynyl-labelled (deoxy)uridine (5-EdU & 5-EU) and the subsequent detection of the ethynyl-functionalised DNA/RNA by covalent conjugation of small-sized fluorescent azides.
5-EdU & 5-EU-based nucleic acid labelling;
• is non-destructive and therefore suitable for mulitparametric analysis (no harsh permeabilisation & nucleic acid denaturation required).
• provides a significantly faster detection procedure (∼2h instead of > 4h). • is compatible with flow cytometry &
microscopy
• is compatible with many cell lines and organisms
CytoTell™ – An alternative to CFSE
Among all the existing fluorescent dyes, CFSE is the preferred cell proliferation indicator that is widely used for live cell analysis. However, there are a few major drawbacks associated with the use of CFSE for monitoring cell proliferation.
• CFSE cannot be used with GFP labelled cells or FITC channel labelled antibodies • CFSE is highly toxic to cells since it
indiscriminately reacts with all amino groups.
• CFSE has slow response and is inconvenient to use. The CFSE fluorescence intensity of the 2nd
generation cells is decreased more than 10 fold from the 1stgeneration. You would
have to wait for another generation to start the cell proliferation analysis;
• Medium removal is required for analysis by Flow Cytometry as CFSE reacts with medium components.
The CytoTell™ Dyes have been developed to eliminate the above CFSE limitations and are available with Red, Green and Blue spectra for ultimate flexibility
CytoTell™ dyes can also be used for long term tracking of labelled cells and are compatible for use with standard formaldehyde-containing fixatives and saponin-based permeabilisation buffers for additional/antibody based staining. CytoTell™ Dyes provide;
• Much faster response to cell proliferation than CFSE.
• Greater convenience than CFSE. • Increased sensitivity over CFSE.

Cytokines are signalling molecules that have critical roles in many biological processes such as cellular growth, differentiation, gene expression, migration, immunity and inflammation. Their malfunction leads to many diseases, including arthritis, acute and chronic liver disease, inflammatory bowel disease, cardiac-related diseases and cancers. Cytokines are commonly working together in a biological or disease process,
therefore, the comprehensive analysis of the expression of multiple cytokines is important in revealing the mechanisms underlying many biological functions and diseases.
The Human Cytokine ELISA Plate Array from Signosis is available with chemiluminescent or colourimetric detection that allows you to monitor the abundance of 31 human cytokines simultaneously. This fast and sensitive assay can be used for quantitative comparison of these cytokines among different samples.
The 96-well white plate is divided into 3 or 4 sections, and each section has 4 or 3 columns for one sample. In each section, 31 specific cytokine capture antibodies are coated on 31 wells and one blank. The sample can be cell culture supernatants, cell lysates, tissue homogenates, serum, or plasma samples among others.
