Peeking Under the Hood of

Cellular Signaling Machinery with Targeted Proteomics Assays

In order to understand the functional nature of cells, it is imperative to study the regulatory aspects of proteins. Proteomic profiling methods are aimed at quantitatively measuring the differences in protein expression across samples. However, proteomic experiments are often plagued by poor reproducibility and failure to consistently observe desired analytes. These shortcomings make it difficult to take advantage of the many tools and methods developed to analyze genomic and transcriptomic data. To address these issues, scientists have begun to develop completely targeted proteomic assay approaches that provide information about several biological disciplines using automated biochemical platforms that will ensure consistently reproducible data for longitudinal studies.

Recently, several protein and peptide sample preparation workflows were automated using the Agilent AssayMAP Bravo platform, which reduced sample preparation variability, provided a clear path for sample processing scalability, and enabled customized workflow development without the need for in-house automation expertise. Additionally, the development of a high-capacity phosphopeptide enrichment cartridge and integrating its use with the AssayMAP system should decrease overall workflow times while simultaneously increasing reproducibility.

Our panelists for this webinar will discuss their experiences with the phosphopeptide enrichment cartridge and automated AssayMAP sample preparation platform, as well as describe some new assay methodologies and preliminary study results.

Our first presenter, Jason D. Russell, Ph.D., LC/MS scientist for Agilent Technologies, will provide a brief introduction to the development of an Fe(III)-NTA phosphopeptide enrichment cartridge for the AssayMAP platform, as well as describe Agilent’s approach toward automated workflow solutions for LC/MS applications.

The second presentation will be given by Jacob D. Jaffe, Ph.D., director of the LINCS Proteomic Characterization Center for Signaling and Epigenetics at the Broad Institute. Dr. Jaffe will discuss his recent work on drug perturbational profiling of cells and the benefits of scalability via automation, including phosphopeptide enrichment, has yielded for his research.
